Polymorphic’s state-of-the-art production facility is ideally suited for high throughput DNA sequencing. Our DNA sequencing service starts with customer-provided templates such as purified plasmids or purified PCR products. We use high throughput liquid handling robotics optimized for automated reaction setup and dye terminator removal. Polymorphic DNA Technologies employs automated high throughput capillary electrophoresis DNA sequencing instruments. Read lengths vary depending on the template provided. Results can be provided electronically by email, by secure download from our website, or CD.

Polymorphic offers a high throughput DNA sequencing service for $2.50 per sample. Our minimum order size is 384 samples.

Please use these instructions as a general guideline for submitting DNA samples to Polymorphic DNA Technologies for high-throughput sequencing services. High-throughput Sequencing Order Specifications:
1. 384 sample minimum
2. 96 well or 384 well plates only. No tubes.
3. Universal Primers available free of charge (T7, M13F, M13R, SP6, T3)

  • Download the Sequencing Order Form and list all appropriate contact and billing information. Be sure to include purchase order or credit card information. Download our 96 well form. Download our 384 well form.
  • Please provide template ID, corresponding primer information, and sample details in the designated areas on the order form.
  • If you are using one primer for every sample, please send 1 tube at a concentration of 5uM. Send at least 5 picomoles for each sequencing reaction. If your order contains more than one sequencing primer, please create a “mirror plate” that exactly matches the layout of your template plate. We need at least 15ul per well of each individual primer OR 100ul per tube.
  • Plasmid DNA or PCR samples should be purified by a commercially available column or gel filtration method.
  • 10nM Tris or DI water are suitable buffers

DNA concentration requirements:
Plasmid DNA –
Minimum 15 microliters of DNA per sequencing reaction at a concentration of 0.04ug/ul
PCR Products – 15ul of product at a concentration of 0.04ug/ul

  • Samples should be provided in 96-well thermocycler plates. Skirted plates are ideal for our automation systems.
  • Submit the completed order form as an attached file in an email to: info@polymorphicdna.com.

Sample Shipping Instructions:

  • Please seal plates tightly for shipping. Strip caps work best, but you may also use a strong adhesive seal or thermoseal . Here are some plate and cap combinations that work well:
    Abgene: www.abgene.com, 96-well skirted PCR plates (AB-1000) and strip caps AB-0265
    Eppendorf: www.eppendorf.com, twin.tec PCR plates and capstrips (listed under PCR consumables)
  • Make sure that plates are protected from movement during shipping. Please include a copy of the customer information section of the order form in your shipment.
  • Samples can be shipped on dry ice, with cold packs, or at ambient temperatures. Over-night service is suggested.

  • Samples are received Monday-Friday, 8am-5pm Pacific Time. No deliveries will be received on Saturdays and Sundays.

Suggestions for High-throughput DNA Sequencing Success:

  • Make sure your samples are accurately concentration normalized across the plate. Variable concentrations lead to variable DNA Sequencing signal.
  • 40ng/ul is the optimal concentration on our AB 3730xl DNA Sequencers. Our Big Dye 3.1 reactions are automated and our process is optimized for this amount of DNA.
  • Please include the size of your insert or the length of your PCR product on the order form. This will help us avoid any project set-up delays.
  • Please don’t mix PCR products and plasmid DNA on one plate. They typically run under different DNA sequencing conditions. If you have a mix of samples, please send separate plates.
  • Your data will be delivered by an FTP link. You will receive an e-mail from Polymorphic DNA Technologies’ customer service. Please copy and paste the link into your browser to access you data. The data will be available on the site for 5 business days. Hard copies are available and can be sent via US mail.
