Shotgun Library Construction & Sequencing
Shotgun library construction and sequencing can begin with a variety
of samples such as cosmid, BAC, and YAC DNA. After a customer
consultation to determine the goals and needs of a specific project,
Polymorphic offers the following shotgun library service(s):
Construction:
- Plasmid
DNA purification and/or quality analysis of in-coming DNA
- Random
shearing to produce a population of DNA fragments within 1-3

Sequencing:

Upon conclusion of the project, the customer is provided with
a report including the original electropherograms, all primer
sequence information, and the full consensus sequence. All plasmid
DNA and primers are delivered.

High-quality
mRNA is synthesized into cDNA using oligo dT primers. Once both
strands are synthesized and annealed, the double stranded cDNA
is directionally cloned into an expression vector.
