SNP Genotyping by Sequencing - Custom assays for any SNP

Process: We use our amplicon sequencing methods to produce small PCR products that serve as templates for bi-directional Sanger sequencing. For this genotyping application, we design smaller amplicons and we optimize the conditions to maximize sequencing accuracy at the target SNP site. The reporting focuses on the SNP(s) of interest, although the sequences of the flanking regions are also available.

Sample QC. Prior to genotyping, we can perform real-time PCR to accurately determine the concentration of functional DNA in human genomic samples. We run all samples in duplicate and we report the final DNA quantities measured.

Assay validation. We can apply each assay to a library of Coriell DNA samples. This library of samples is from an ethnically diverse population and contains trios and replicates useful for assay assessment.
